تعیین چندشکلی ژن های POU1F1 و STAT5A و ارتباط آنها با صفات تولید شیر در گاو

چکیده

ژن های کاندیدا برای صفات خاص، ژن های توالی یابی شده ای هستند که فعالیت بیولوژیکی آنها شناخته شده است. در این میان نقش ژن های POU1F1 و STAT5A روی صفات تولید شیر گاو در تحقیقات مورد بررسی قرار گرفته است. ژن POU1F1 روی نسخه برداری پرولاکتین و هورمون رشد تأثیر دارد و ژن STAT5A به عنوان تنظیم کننده اصلی فعالیت هورمون رشد در سلول های هدف و مبدل سیگنال داخل سلولی پرولاکتین عمل می کند. از آنجا که این ژن ها برای توسعه و تکامل سیستم پستانی و تولید شیر مهم هستند، هدف از این تحقیق بررسی چندشکلی جایگاه آنها و ارتباطشان با ارزش اصلاحی صفات تولید شیر در گاوهای براون سوییس بود. بدین منظور 90 رأس گاو شیری بطور تصادفی خونگیری شدند. استخراج DNA از خون تام به روش نمکی تغییر یافته انجام گرفت و واکنش زنجیره ای پلی مراز (PCR) جهت تکثیر قطعات مورد نظر انجام شد. قطعات تکثیر شده با آنزیم های محدودالاثر اختصاصی هضم و ژنوتیپ ها تعیین شدند. فراوانی ژنی و ژنوتیپی، شاخص های هتروزیگوتی، تعداد آلل مؤثر و واقعی، برآورد ارزش اصلاحی صفات تولیدی و ارتباط ژنوتیپ با ارزش اصلاحی صفات، در این تحقیق مورد بررسی قرار گرفت. فراوانی آلل های A و C ژن های POU1F1 و STAT5A به ترتیب 455/0 و 489/0 و آلل های B و T این جایگاه ها به ترتیب 545/0 و 511/0 برآورد شد. جمعیت مورد مطالعه در هر دو جایگاه در تعادل هاردی واینبرگ بود. ارتباط معنی داری بین جایگاه های مورد مطالعه و ارزش اصلاحی صفات پیدا نشد، اگرچه آلل POU1F1*B گرایش به تولید شیر بیشتر و POU1F1*A تمایل به تولید بالاتر درصد چربی شیر و پروتئین نشان دادند.

Specific trait candidate genes are sequenced genes with known biological activity. The effects of POU1F1 and STAT5A on milk production traits have been studied in several studies. POU1F1 affects on transcription of prolactin and growth hormone gene, as well as, STAT5A is known as a main mediator of growth hormone action on target genes and intracellular mediator of prolactin signaling. Since these genes are essential for development of mammary system, the aim of this study was to determine association of their polymorphism with milk production breeding values in Brown Swiss cattle. Blood of ninety milking cow were randomly obtained. DNA was extracted from whole blood using modified salting out method, then the desired fragments were PCR amplified and digested by specific restriction endonuclease enzymes. Gene and genotype frequencies, heterozygosity indexes, the real and effective allele number were calculated by PopGene software; and the breeding values of production traits were estimated by DFREML. SAS software was used to analyze association between genotypes and breeding values. The frequency of 'A' and 'C' alleles of POU1F1 and STAT5A were 0.455 and 0.489, respectively. This population was in hardy-weinburg equilibrium for both loci. There was no significant association between genotypes and breeding values, although POU1F1*B tended to produce higher milk and POU1F1*A showed higher fat and protein percent.
