بررسی تنوع ژنتیکی جایگاه ژن DGAT1 و میزان ارتباط آن با تولید شیر در جمعیت گاو های هلشتاین ایران

چکیده

در سال های اخیر در طرح های نقشه یابی QTL مشخص شده است که یک ژن کاندید بالقوه (DGAT1) در انتهای سانترومری کروموزوم شماره 14 برای درصد چربی و تولید شیر وجود دارد. ژنDGAT1 با کد کردن آنزیم دی آسیل گلیسرول اسیل ترانسفراز (DGAT) نقش اصلی را در سنتز تری گلیسیرید و چربی شیر دارد. ایجاد یک جهش تک نوکلئوتیدی باعث تبدیل گوانین به آدنین در مولکول DNA ، و منجر به جایگزینی آلانین به جای لیزین در آنزیم DGAT می‏گردد. وجود این جهش اثر زیادی روی تولید و ترکیب شیر دارد. در این پژوهش 398 نمونه خون از گاوهای هلشتاین استانهای تهران و اصفهان جمع آوری گردید، سپس استخراج DNA با استفاده از روش نمکی انجام پذیرفت. با انجام واکنش PCR یک قطعه 411 جفت بازی از اگزون شماره 8 این ژن تکثیر گردید. تعیین ژنوتیپ جمعیت با استفاده از تکنیک PCR-RFLP انجام شد. بر اساس نحوه برش آنزیم CfrI سه نوع ژنوتیپ KK ،KA ،AA مورد انتظار بود که، افراد KK تیپ وحشی هستند. افراد KA دارای یک آلل جهش یافته (A) و افرادAA دارای دو آلل جهش یافته هستند. نتایج این تحقیق نشان داد که 36 فرد دارای ژنوتیپ KK ، 226 فرد دارای ژنوتیپ KA و 136 فرد دارای ژنوتیپ AA می باشند. بنابراین فراوانی آلل K برابر37/0 و فراوانی آلل A برابر با 63/0 برآورد گردید. همچنین میزان اثر این ژن روی تولید شیر 305 روز در سطح1/0 درصد معنی دار بدست آمد

The results of multiple QTL mapping design in recent years indicated quantitative trait loci for milk fat percentage and milk yield traits identified in BTA14. DGAT1 gene coding diacylglycerol-acyltransferase enzyme, has a main role in the triglyceride synthesis and finally fat milk synthesis. A transition mutation in this gene results substitution of guanine by adenine in the gene causing substitution of lysine by alanine in diacylglycerol-acyltransferase enzyme. In this research, 398 blood samples were collected from the farms Tehran and Esfahan provinces. Using PCR reaction, 411 bp fragment of exon 8 DGAT1 gene was amplified. Finally, genotyping of the population was performed using RFLP- PCR technique. Three genotypes were detected. KK individuals with no mutant allele, AA individuals with two mutant alleles and KA individuals with one mutant allele. 36, 226 and 136 individuals had KK,KA and AA genotypes, respectively. Frequencies of DGAT1 alleles (K and A) were estimated 0.37 and 0.63, respectively. The effect of DGAT1 gene on milk305 trait was significant (P=0.001).
