چکیده

اين آزمایش به منظور مطالعه اثرات پرتو ميكروويو بر تجزيه‌پذيري ماده خشك و پروتئين خام و حقيقي، قابليت هضم برون‌تني پروتئين خام، تركيبات ضد تغذيه‌اي (گلوكوسينولات و اسيد فايتيك) و تركيبات شيميايي كنجاله منداب اصلاح شده انجام شد. تیمارهای آزمایشی شامل کنجاله منداب اصلاح شده عمل‌آوری نشده و کنجاله منداب اصلاح شده عمل‌آوری شده با پرتو میکروویو با قدرت 800 وات به مدت 2، 4 و 6 دقیقه بودند. ميزان تجزيه‌پذيري ماده خشك و پروتئين خام و حقيقي به روش كيسه‌هاي نايلوني اندازه‌گيري شد. مقدار 6 گرم از هر تیمار به مدت صفر، 2، 4، 8، 16، 24 و 48 ساعت در شكمبه سه رأس گاو نر تالشي انكوباسيون شد. میزان پروتئين حقيقي با روش برادفورد و نحوه تجزيه شدن آن در زمان‌هاي مختلف انكوباسيون در شكمبه با روش الكتروفورز SDS-PAGE تعيين شد قابليت هضم برون‌تني پروتئين خام به روش آنزيمي سه مرحله‌اي تعیین شد. عمل‌آوري با پرتو ميكروويو سبب كاهش اسيد فايتيك و گلوكوسينولات‌هاي کنجاله منداب اصلاح شده به صورت روند خطی و درجه دو شد. پرتوتابی با میکروویو به مدت 2، 4 و 6 دقیقه اسید فایتیک کنجاله منداب اصلاح شده را به ترتیب به میزان 13، 30 و 51 درصد نسبت به کنجاله منداب عمل‌آوري نشده کاهش داد. مقدار گلوکوسینولات‌های کنجاله منداب پرتوتابی شده با میکروویو به مدت 6 دقیقه به میزان 67 درصد نسبت به کنجاله عمل آوری نشده کاهش یافت. پرتو ميكروويو كاهش بخش سريع تجزيه، نرخ ثابت تجزيه و تجزيه‌پذيري مؤثر ماده خشك و افزايش بخش كند تجزيه ماده خشك و پروتئين خام و حقيقي کنجاله منداب اصلاح شده را سبب شد. قابليت هضم برون‌تني پروتئين خام كنجاله‌ منداب اصلاح شده با عمل‌آوری با میکروویو تا 4 دقيقه به طور معني‌دار افزايش يافت. الكتروفورز پروتئين‌ كنجاله منداب اصلاح شده نشان داد كه عمده پروتئين در آنها شامل ناپين (آلبومين S2) با دو زيرواحد و كروسيفرين (گلوبولين S12) با چهار زيرواحد بود. تجزيه‌هاي الكتروفورز پروتئين کنجاله منداب اصلاح شده نشان داد كه در کنجاله منداب اصلاح شده عمل‌آوري نشده چهار زيرواحد كروسيفرين؛ در كنجاله کنجاله منداب اصلاح شده عمل‌آوري شده، چهار زيرواحد كروسيفرين و زيرواحدهاي ناپين بخش عمده پروتئين عبوري را تشكيل دادند. نتايج اين پژوهش نشان داد كه عمل‌آوري با ميكروويو سبب كاهش تجزيه‌پذيري پروتئين خام و حقيقي در شكمبه، افزايش قابليت هضم برون‌تني پروتئين خام و كاهش مقدار تركيبات ضد تغذيه‌اي كنجاله منداب اصلاح شده شد.
