تأثیر تغذیه متوالی و متناوب بتاآگونیست زیلپاترول هیدروکلراید روی بیان ژن گیرنده های بتاآدرنرژیک ماهیچه در بره های نر پرواری
هدف از این آزمایش ارزیابی اثرات سه روش مختلف مصرف زیلپاترول هیدروکلراید به مدت 42 روز آخر دوره پرواربندی روی بیان ژن گیرنده های 1β و 2β آدرنرژیک در بره های نر پرواری لری-بختیاری بود. تغذیه متوالی زیلپاترول (روزانه)، تغذیه متناوب و به صورت یک روز در میان (1on 1 off) و تغذیه متناوب و به صورت دو روز مصرف و دو روز استراحت (2on 2off) به عنوان روش های مختلف تغذیه به کار برده شدند. تعداد 32 راس بره نر با میانگین وزن اولیه 7/4± 44 کیلوگرم براساس وزن اولیه به 4 گروه (8n=) تقسیم و تا آخر دوره پرواربندی با جیره حاوی 14 درصد پروتئین و 2/0 میلی گرم زیلپاترول به ازای هر کیلوگرم وزن زنده بره ها تغذیه شدند. جیره پایه بدون زیلپاترول به عنوان گروه شاهد در نظر گرفته شد. به منظور تعیین بیان ژن گیرنده های بتاآدرنرژیک، سه مرحله نمونه گیری به روش بیوپسی از ماهیچه ران از 3 راس بره در هر تیمار در روزهای صفر، 21 و 42 آزمایش انجام شد و نمونه ها بلافاصله در نیتروژن مایع قرار داده شدند. در آزمایشگاه پس از استخراج RNA کل از بافت ماهیچه، cDNA با استفاده از آنزیم رونوشت بردار معکوس سنتز شد. پس از طراحی آغازگرها، RT-PCR برای نمونه های cDNA ، با استفاده از دستگاه iQ5 BioRad انجام شد. نتایج این تحقیق نشان داد که اثرات اصلی و اثر متقابل مدت زمان مصرف و روش مصرف زیلپاترول روی بیان نسبی ژن گیرنده1 β و2β آدرنرژیک معنی دار نبود. همچنین در مقایسه تیمارها با گروه شاهد نیز تفاوت معنی داری در سطح 05/0 مشاهده نشد. بنابراین تغذیه متناوب زیلپاترول هیدروکلراید بیان ژن گیرنده های بتاآدرنرژیک را تحت تأثیر قرار نداد.

تاثیر جایگزینی سطوح مختلف کاه گندم با برگ خرما بر عملکرد و سلامتی برههای بلوچی
هدف از این مطالعه، ارزیابی تاثیر تغذیه جیره های حاوی برگ خرما بر رشد و عملکرد بره های بلوچی بود. تعداد 24 رأس بره ماده بلوچی با میانگین سنی 4 تا 5 ماه و وزنی 9/3 ± 48/20 کیلو گرم، با یک جیره کل مخلوط شامل 39 درصد علوفه و 61 درصد کنسانتره تغذیه شدند. بخش علوفهای جیرههای آزمایشی شامل 1)کاه گندم 24 %، 2) کاه گندم 16% و برگ خرما 8%، 3) کاه گندم 8% و برگ خرما 16% و 4) برگ خرما 24 % بود. تمامی برهها با یک کنسانتره یکسان تغذیه شدند. میانگین مصرف خوراک روزانه بره های تغذیه شده با جیره های مختلف به ترتیب 856، 917، 983 و 1033 گرم در روز برای جیره 1 تا 4 بود. اختلاف بین میانگین مصرف خوراک روزانه در جیره های مختلف معنی داربود (05/0P

اثر افزودنی باکتریایی و مواد جاذب رطوبت بر قابلیت تخمیر و ترکیب مواد مغذی سیلاژ تفاله چغندرقند با استفاده از سیلوهای آزمایشگاهی
هدف از این مطالعه بررسی نقش افزودنی میکروبی تولید کننده اسید لاکتیک و همچنین جاذب های مختلف در کاهش پس آب و بهبود ویژگی های شیمیایی و تخمیری سیلاژ تفاله چغندرقند تازه بود. در آزمایش اول تفاله چغندر بصورت خالص یا به صورت مخلوط شده با 5 درصد کاه یا 5 درصد پیت بمدت 90 روز سیلو شد. در آزمایش دوم تفاله چغندر با افزودنی میکروبی، پیت و یا مخلوط آنها سیلو شد. غلظت ماده خشک و دیواره سلولی در سیلاژهایی که کاه یا پیت دریافت کرده بودند بیشتر از تیمار شاهد بوده و پس آب تولیدی کاهش پیدا کرد (01/0P

اثر سطوح پروتئین و اسیدهای آمینه گوگرد دار جیره آغازین بر عملکرد و بافت شناسی ژژنوم جوجه های گوشتی
آزمایشی به منظور بررسی اثر پروتئین و اسیدهای آمینه گوگرد دار قابل هضم جیره آغازین بر عملکرد و بافت شناسی ژژنوم جوجههای گوشتی در قالب طرح کاملا تصادفی به ترتیب فاکتوریل3×2 انجام شد. 300 قطعه جوجه گوشتی نر یک روزه سویه راس به 30 گروه 10 قطعه ای تقسیم شدند. تیمارهای آزمایشی شامل دو سطح پروتئین قابل هضم 5/19 و 5/21 درصد و سه سطح اسیدهای آمینه گوگرد دار قابل هضم (DSAA) 94/0، 02/1 و 1/1 درصد جیره بودند. افزایش درصد DSAA ، مصرف خوراک و ضریب تبدیل را در دوره آغازین افزایش داد اما بر افزایش وزن در این دوره تأثیری نداشت. با افزایش درصد DSAA در دوره آغازین، افزایش وزن روزانه و مصرف خوراک در دوره پایانی و کل دوره افزایش یافت اما بر ضریب تبدیل تأثیری نداشت. با افزایش سطح DSAA در دوره آغازین، اجزای لاشه و وزن نسبی قسمت های مختلف روده کوچک در10 و42 روزگی اختلاف معنیداری را نشان نداد اما چربی محوطه شکمی در 42 روزگی کاهش یافت. با افزایش سطح پروتئین قابل هضم، ضخامت ماهیچه در ژژنوم در 10 روزگی افزایش یافت. با افزایش درصد DSAA ،طول و عرض پرز در ناحیه ژژنوم در 10 روزگی افزایش یافت.سطوح پروتئین قابل هضم و DSAA تأثیری بر طول نسبی اجزای روده کوچک در 10 و 42 روزگی نداشت.

اثر سطوح مختلف عصاره روغنی گل همیشه بهاربرعملکرد، فراسنجههای خونی و پاسخ ایمنی در جوجه های گوشتی درگیر شده با تتراکلرید کربن
این مطالعه به منظور بررسی اثر سطوح مختلف عصاره روغنی گل همیشه بهار بر عملکرد، فراسنجه های خون و سیستم ایمنی جوجه های گوشتی طی یک دوره 42 روزه انجام گرفت. تعداد 200 قطعه جوجه گوشتی نر سویه راس 308 در یک طرح کاملاَ تصادفی با پنج تیمار و چهار تکرار و 10 پرنده در هر تکرار مورد آزمایش قرار گرفتند. تیمارهای آزمایشی شامل گروه کنترل منفی (فاقد افزودنی عصاره گل همیشه بهار و تتراکلریدکربن)، کنترل مثبت (یک میلی گرم بر کیلوگرم وزن بدن) و سه سطح عصاره گل همیشه بهار (150، 300 و 450 میلی گرم بر کیلوگرم خوراک) به همراه تتراکلریدکربن و جیره پایه بود. از روز 21 تا 30 آزمایش، هر دو روز، تتراکلریدکربن به عنوان عامل درگیرکننده ی کبد، به صورت داخل صفاقی و در طول این مدت به گروه کنترل منفی یک میلی گرم در کیلوگرم وزن بدن محلول کلرید سدیم 9/0% تزریق شد. صفات مربوط به عملکرد اختلاف معنی داری را از نظر آماری نشان ندادند. جوجه های تغذیه شده با تترا کلرید کربن بالاترین وزن نسبی کبد را در مقایسه با تیمارهای دریافت کننده ی عصاره گل همیشه بهار داشتند. وزن نسبی تیموس و سطح ایمنوگلوبولین Gدر تیماری که علاوه بر تتراکلریدکربن، سطح 300 میلی گرم عصاره گل همیشه بهار را دریافت کرده بودند، به صورت معنی داری بالاتر بود. فراسنجه های خون (SGOT وSGPT) که در اثر تزریق تتراکلریدکربن، سطح شان در خون افزایش یافته بود، عصاره گل همیشه بهار توانست اثرات منفی ناشی از این ماده سمی را تقلیل دهد. سطوح کلسترول و تری گلیسرید در تیمار ۱۵۰ و ۴۵۰ میلی گرم در کیلوگرم خوراک عصاره همیشه بهار نسبت به گروه کنترل مثبت کاهش معنی دار داشت. نتایج نشان داد عصاره گل همیشه بهار می تواند به عنوان محافظت کننده کبد در مواجهه با سموم مختلف در تغذیه طیور مورد استفاده قرار گیرد.

بررسی عوامل مؤثر بر صفت طول عمر اقتصادی در گوسفند کردی
به منظور بررسی عوامل مؤثر بر صفت طول عمر اقتصادی گوسفند کردی خراسان شمالی از رکوردهای ثبت شده ایستگاه پرورش و اصلاح نژاد گوسفندکردی طی سال های 1369 تا 1391 که شامل 7469 بره حاصل از 187 پدر و 2258 مادر بود، استفاده گردید. ترسیم جداول طول عمر تولیدی از رویه Life table نرم افزار SAS نسخه 1/9 صورت گرفت. عوامل ثابت سال و فصل بر طول عمر حیوان اثر معنی داری داشت. نتایج نشان داد که حدود 99 درصد از میش های داشتی به دلایل غیر از سن از گله حذف گردیده اند. بیشترین درصد حذف در میش ها مربوط به بیماری (1/33%) و کمترین درصد حذف در جنس نر مربوط به مسمومیت (11/0%) می باشد. تعداد میش های حذف شده از گله با بالا رفتن سن کاهش می یابد، اما حذف میش های بالای 4 سال در مقایسه با میش های جوان افزایش یافت. زنده مانی میش هایی که زایش تک قلو داشتند، بیشتر از میش هایی است که زایش دو قلو داشتند و میش های دارای 1 بره نسبت به میش های دارای 2 و 3 بره، درصد زنده مانی بیشتری را نشان دادند. نتایج نشان داد که میش هایی که در تمام زایش ها یک بره داشتند دارای بالاترین درصد ماندگاری هستند، درحالی که در میش هایی دارای دو بره، درصد زنده مانی کمتر مشاهده شد. اگر چه سیاست های کلی در دام های سبک به سمت افزایش میزان دوقلوزایی می باشد، اما نتایج نشان داد تأکید بر دوقلوزایی موجب حذف میش ها در سنین پایین و کاهش طول عمر اقتصادی خواهد شد. برای بهبود ماندگاری می توان شرایط مدیریتی و محیطی گله را بهبود بخشید.

استفاده از روش PCR رقابتی و Real-time PCR مطلق به منظور کمی سنجی جمعیت باکتری تولید کننده بوتیرات: بوتیریویبریو فیبری سالونس
سویه های باکتری بوتیریویبریو فیبری سالونس به عنوان عمده ترین گروه باکتری های تولید کننده بوتیرات در دستگاه گوارش بسیاری از حیوانات و همچنین انسان شناخته شده اند. در این مطالعه دو تکنیک مبتنی بر DNA شامل PCR رقابتی و Real-time PCR به منظور شمارش باکتری های گونه بوتیریویبریو فیبری سالونس استفاده شدند. در ابتدا آغازگرهای اختصاصی برای منطقه 16S rDNA باکتری بوتیریویبریو فیبری سالونس به منظور تکثیر یک قطعه 213 جفت بازی استفاده شدند. یک قطعه رقابتگر با تفاوت طول 50 جفت باز بیشتر ساخته شد و در ناقل پلاسمیدی pTZ57R/T همسانه سازی شد. کمیت رقابتگر پلاسمیدی با استفاده از نانودراپ اسپکتروفتومتری سنجیده شد و به صورت سریالی رقیق سازی شد. رقت های حاصل به همراه DNA استخراج شده از مایع شکمبه بطور همزمان تکثیر شدند. به منظور کمی سازی محصولات PCR پس از عکس برداری از ژل آگارز و استفاده از نرم افزار ImageJ ، مقادیر تکثیر شده از DNA هدف در مقابل مقادیر تکثیر شده از رقابتگر به صورت لگاریتمی ترسیم شدند. از ضریب تبیین (R2) به عنوان معیاری جهت ارزیابی دقت تکنیک استفاده گردید. برای توسعه Real-time PCR، قطعه 213 جفت بازی تکثیر شده و همسانهسازی شده در ناقل پلاسمیدی pTZ57R/T به منظور رسم منحنی استاندارد استفاده شد. نتایج نشان دادند که هر دو روش قابلیت استفاده برای شمارش باکتری های جنس بوتیریویبریو فیبری سالونس را دارا هستند و بنابراین می توانند به عنوان ابزارهای مناسب در تحقیقات مرتبط با این باکتری مورد استفاده قرار بگیرند.

بررسی میزان و نوع آلودگی قارچی خوراک دام درگاوداری های شیری شهر یزد
آلودگی خوراک و غذای دامها به قارچ های سمی شایع موجود در هوا و محیط حایز اهمیت میباشد. خوراک دام آلوده به قارچهای مولد آفلاتوکسین منجر به اختلال در چرخه سلامت دام، شیر و افراد مصرف کننده فراورده های دامی می گردد. این مطالعه با هدف بررسی میزان و نوع آلودگی قارچی خوراک دام درگاوداری های شیری شهر یزد انجام شد. این تحقیق توصیفی- مقطعی در تابستان 91 بر روی خوراک دام 23 گاوداری شیری در شهر یزد انجام شد. نمونه گیری از خوراک دام بصورت تصادفی ساده صورت گرفت. نمونه ها بر روی محیط کشت سابورو دکستروز آگار کشت داده و نوع قارچهای آلوده کننده تعیین و تعداد کلنی شمارش و نتایج بدست آمده با نرم افزار 16SPSS و با استفاده از آزمون های کای اسکور و آزمون من ویتنی مورد تجزیه و تحلیل آماری قرار گرفت. یافته های تحقیق حاضر نشان داد که قارچهای هیفومیست سیاه و قارچهای ساپروفیت از قارچهای شایع آلوده کننده خوراک دام بودند. همچنین کاه بیشترین آلودگی به قارچهای سمی مولد آفلاتوکسین را داشت. از بین خوراک های دام به ترتیب تعداد کلنی های قارچی رشد کرده روی سیلاژ ذرت (42600) و کنسانتره (40600) در هر گرم خوراک دام، بالاترین میانگین آلودگی را دارا بودند. کنترل آلودگی قارچی خوراک مصرفی در گاوداری ها به قارچ ها، بهترین روش برای جلوگیری از آلودگی شیر و فرآورد های آن به آفلاتوکسین است که به بهبود سلامت جامعه نیز کمک می کند.

ارزیابی ترکیب شیمیایی، گوارش پذیری وکیفیت پروتئین علوفه تاج خروس در دو مرحله برداشت
در این پژوهش ترکیبات شیمیایی ، قابلیت هضم، اجزای پروتئین بر اساس CNCPS و مواد ضد مغذی علوفه حاصل از دو رقم گیاه تاج خروس که در شرایط یکسان کشت وطی دو مرحله متوالی برداشت گردید، در قالب طرح کاملاً تصادفی مورد بررسی قرار گرفت. نتایج نشان داد که میانگین خاکستر خام، الیاف نامحلول در شوینده خنثی، الیاف نامحلول در شوینده اسیدی، لیگنین، کلسیم، فسفر، منیزیم و پتاسیم به ترتیب؛ 44/15، 85/40، 90/27، 06/5، 55/1، 25/0، 28/0 و 48/1 درصد در ماده خشک بود. همچنین میانگین ماده خشک گوارش پذیر، ماده آلی گوارش پذیر و ماده آلی گوارش پذیر در ماده خشک، با روش برون تنی (آزمایشگاهی) به ترتیب 19/60، 16/58 و 09/49 درصد محاسبه گردید. از نظر گوارش پذیری تفاوت بین ارقام مرحله برداشت و نیز اثر متقابل رقم در مرحله برداشت معنیدار نبود. میزان پروتئین خام در دو رقم علوفه تاج خروس 72/14 درصد ماده خشک و مقادیر پروتئین حقیقی، پروتئین محلول و بخشهای A، B1، B2، B3 و C به ترتیب برابر 52/59، 71/42، 48/40، 24/2، 19/34، 09/16 و 01/7 درصد از پروتئین خام و نیتروژن غیر پروتئینی 55/94 درصد از کل پروتئین محلول محاسبه گردید. انرژی قابل متابولیسم 56/2 مگاکالری در کیلوگرم ماده خشک برآورد شد. همچنین مقادیر نیترات و اگزالات، به طور میانگین در دو رقم علوفه مزبور به ترتیب 34/0 و 37/5 درصد در ماده خشک اندازهگیری شد. به طور کلی نتیجه گرفته شد که با توجه به ارزش تغذیه ای و ضرایب هضمی بالای تاج خروس و همچنین مواد ضد مغذی پایین تر از حد خطرناک برای دام در آزمایش حاضر، می توان از آن به عنوان علوفه ای با کیفیت در تغذیه نشخوار کنندگان استفاده نمود.

تاثیر جو و آنزیم بر عملکرد، لاشه، فعالیت آنزیمی و فرآسنجههای گوارشی جوجههای گوشتی
آزمایشی برای بررسی اثر جیره جو مکمل شده با آنزیم در مقایسه با ذرت بر صفات تولیدی و مشخصات فیزیولوژیک جوجه های گوشتی با استفاده از 375 قطعه جوجه یک روزه مخلوط سویه تجاری راس 308 در قالب طرح کاملا تصادفی با 3 تیمار و 5 تکرار به مدت 42 روز اجرا گردید. صفات مورد بررسی شامل عملکرد رشد در دوره پرورش و اندازهگیری صفات لاشه ، فعالیت آنزیمی لوزالمعده و خصوصیات فیزیکی- شیمیایی محتویات گوارشی ایلئوم در سن 42 روزگی بودند. تیمارها شامل شاهد، جو و جو با آنزیم بودند. نتایج نشان داد تاثیر جیره جو با و بدون آنزیم بر عملکرد رشد در دورههای آغازین، رشد و کل دوره پرورش و بر خصوصیات لاشه، فعالیت آنزیمی لوزالمعده و مشخصههای فیزیکی- شیمیایی محتویات ایلئومی جوجهها در سن 42 روزگی معنیدار بود (01/0P

اثر تفالههای لیمو، انگور و سیب بر عملکرد، صفات لاشه، خصوصیات دستگاه گوارش، مورفولوژی روده و صفات ایمنی در جوجههای گوشتی
این آزمایش جهت ارزیابی اثرات استفاده از تفالههای لیمو، انگور و سیب بر عملکرد، صفات لاشه، خصوصیات دستگاه گوارش، مورفولوژی روده و صفات ایمنی در جوجههای گوشتی انجام شد. تعداد 240 قطعه جوجه گوشتی سویه راس- 308 در 5 تیمار، 4 تکرار و 12 قطعه جوجه در هر تکرار در قالب طرح کاملاً تصادفی مورد آزمایش قرار گرفتند. گروههای آزمایشی شامل: 1) شاهد (بدون استفاده از تفاله)، 2) حاوی 150 میلیگرم بر کیلوگرم ویتامین E (به عنوان شاهد مثبت)، 3) حاوی 3 درصد تفاله لیمو، 4) حاوی 3 درصد تفاله انگور و 5) حاوی 3 درصد تفاله سیب بودند. جیره رشد از سن 11 تا 24 روزگی و جیره پایانی از سن 25 تا 42 روزگی در اختیار جوجهها قرار گرفتند. استفاده از تفالههای انگور و سیب نسبت به شاهد موجب بهبود ضریب تبدیل غذایی گردید )05/0(P. صفات لاشه جوجهها تحت تأثیر جیرههای آزمایشی قرار گرفت )05/0(P

آنالیز ژنتیکی صفات تولیدی گاوهای هلشتاین منطقه ی مدیترانه ای ایران به روش رگرسیون تصادفی و مدل دام
در این مطالعه به منظور برآورد پارامترهای ژنتیکی و فنوتیپی و نیز روند فنوتیپی و ژنتیکی صفات تولیدی گاوهای هلشتاین منطقه ی مدیترانه ای ایران(استانهای اردبیل، همدان، زنجان، آدربایجان شرقی و غربی) از اطلاعات مربوط به8808 گله ی گاو هلشتاین، شامل 105118 رکورد روزآزمون استفاده شد. ویرایش لازم روی تمامی رکورد ها با استفاده از نرم افزارهای Access2010 و Fox Pro 8.0 انجام گرفت. برازش اثرات ثابت موثر برروی رکوردهای روزآزمون با استفاده از رویه GLM نرم افزار SAS انجام گرفت که تاثیر سال- فصل، گله- تاریخِ روزآزمون و استان روی داده های روزآزمون و برای صفات مقدار شیر، درصد چربی و پروتین شیر معنی دار بود. همچنین اثر خطی و درجه دوم سن زایش به عنوان عامل کمکی بر روی هر سه صفت معنی دار بود. برای برآورد روند ژنتیکی و فنوتیپی، از نرم افزار SAS و رویه ی GLM5 استفاده شد. میزان وراثت پذیری برای صفات مقدار شیر، در صد چربی و درصد پروتئین با استفاده از رکودهای روزآزمون به ترتیب، 126/0، 02/0 و059/0 برآورد شد. نتایج نشان می دهند که دقت حاصل از رکورد های روزآزمون با استفاده از روش رگرسیون تصادفی بالاتر از رکودهای تصحیح شده برای 305 روز است و لذا در این مطالعه بر نتایج حاصل از رکوردهای روزآزمون و روش رگرسیون تصادفی تاکید می شود. نتایج حاصل از مدل رگرسیون تصادفی و رکوردهای روزآزمون نشان می دهند که وراثت پذیری تولید شیر، بیشترین و وراثت پذیری درصد چربی کمترین مقدار را به خود اختصاص داده و وراثت پذیری درصد پروتئین در حد وسط قرار گرفته است.

بررسی ژن BMP 15 در گوسفندان افشاری و آمیخته افشاری× برولامرینو
افزایش تعداد بره به ازای هر میش به عنوان راهکاری برای افزایش بهره وری در پرورش گوسفند مورد توجه زیادی قرار گرفته است. صفات تولید مثلی از وراثت پذیری پائینی برخوردار هستند اما امروزه جهش هایی در برخی ژنها از جمله BMP15 یافت شده است که با چند قلوزایی در ارتباط است. هدف از مطالعه حاضر شناسایی چند شکلی و بررسی بیان ژن BMP15 در تخمدان میش های آبستن و غیر آبستن افشاری بود. در این مطالعه از تعداد 35 راس میش افشاری و 45 راس میش آمیخته افشاری× برولا مرینو خون گیری و استخراج DNA به عمل آمد. سه قطعه از ژن BMP 15 به طول های 581، 325 و 857 جفت باز به ترتیب برای ناحیه پروموتر، اگزون یک و دو برای وجود چند شکلی از طریق تعیین توالی محصولات PCR مورد بررسی قرار گرفت. نتایج نشان داد که در پروموتر و اگزون یک هیچ گونه تغییر توالی وجود ندارد اما در اگزون دو، نوکلئوتید 134 دچار تغییر شده و دو ژنوتیپ هموزیگوت CC و هتروزیگوت CA مشاهده گردید. پس از کشتار از 22 راس میش آبستن و 8 راس میش غیر آبستن بافت تخمدان تهیه شد. از نمونه ها RNA کامل استخراج و سپس به cDNA تبدیل شد. نتایج بررسی اختلاف معنی داری از لحاظ بیان ژن BMP15 بین دو گروه آبستن و غیر آبستن و آلل ها نشان نداد. احتمالا این ژن در ارتباط با سایر ژنها و از جمله گیرنده های مربوطه اش در ارتباط با تولید مثل ایفای نقش می کند و به همین دلیل ممکن است بیان آن در مراحل مختلف تولید مثلی مورد نیاز باشد.
